Enhancing Biobank Quality Control: An NGS-QC-PANEL as a Superior Alternative to STR Assays for Large-Scale Sample Authentication

Wubin Qian, Zhenzhen Song, Xiaobo Chen, Sheng Guo

Tumor biobanks store large collections of biospecimens crucial for research, but ensuring the samples remain uncontaminated and correctly identified is challenging. STR profiling, a traditional PCR-based method for sample authentication, has limitations in sensitivity, often missing contamination below 20%.

It also struggles with closely related species like inbred mouse strains and mixed human and mouse samples, common in modern preclinical biobanks. As a result, more accurate and sensitive methods are needed to ensure proper authentication and avoid contamination.

  • How the NGS-QC-PANEL improves sensitivity and accuracy in authenticating human and mouse biospecimens, surpassing traditional STR assays.

  • How biobank quality control improved, reducing contamination and misidentification in over 70,000 samples.

  • Real-world examples of common biobank issues, like model mixing and contamination, and how the NGS-QC-PANEL mitigates these risks for reliable research outcomes.
