Monoclonal Antibody Development

Crown Bioscience has extensive experience developing, characterizing and expressing monoclonal antibodies. Many custom projects for our preclinical clients have culminated in commercially available in vivo grade antibodies that are now offered as part of our Life Science CrownVivoTM and CrownVivoPremiumTM primary antibody collections. Many have been designed for use in detection, neutralization, and in vivo depletion assays. Our most popular host animals are mice and hamsters, with hamsters having the unique advantage of being used in vivo in mouse models without eliciting an immune response against the injected antibody. While specializing in antibodies for early discovery research and in vivo preclinical applications, we can also accommodate antibody development for in vitro diagnostics.

Large Scale In Vitro Monoclonal Antibody and Protein Production

With extensive experience in large scale mammalian cell culture of hybridoma and mammalian cells for production of monoclonal antibodies, Crown is able to support large scale antibody and protein production requests. In cases where projects are still in the proof of concept stage, in vitro production can be performed in roller bottles or shake flasks. Bioreactor production is available at small and large scale.

Rapid Expression of Human Monoclonal Antibody Variants and Recombinant Proteins

Essential in the process of understanding mechanisms of disease, preclinical research enables researchers to create models of disease states and elucidate the biochemical and cellular processes responsible for the development of diseases. We are pleased to offer a range of services to scientists requiring specific antibodies and proteins of the highest purity available for conducting early discovery and preclinical research, including:

The transgene expression system used by Crown has been developed to expedite expression of recombinant proteins and monoclonal antibodies for use in early discovery research, preclinical trials or in vitro diagnostic procedures. It was developed to transfect mammalian cells (CHO or HEK-293) with high efficiency during suspension cell culture in stirred tank bioreactors.

Antibody and Protein Purification

With robust multi-step purification procedures, Crown is able to provide highly purified recombinant proteins and antibodies for use in early discovery research and preclinical trials. A variety of scales from laboratory to commercial manufacturing are available. Development of a purification procedure is based on the final product application and predetermined specifications. In most antibody cases, a robust procedure beginning with protein A chromatography followed by other proprietary chromatography steps result in a highly pure antibody preparation. For in vivo functional studies or preclinical trial materials, procedures resulting in preparations free of aggregates, DNA, viruses, leached protein A and low endotoxins are available.

Final Fill Finish

Conjugates are formulated in stability buffers that increase product shelf life and optimize performance. Final products may be packaged in vials ready for immediate use.

Preparation of F(ab') or F(ab')2 Fragments of Antibodies

We can also support enzyme digestion of antibodies into univalent F(ab') or F(ab')2 fragments for use in immunoassays, immunohistochemistry and in vivo functional assays. Antibody fragments may be adsorbed against serum proteins to remove undesired species cross-reactivity and conjugated to the required reporter molecule.
