Project Design and Immunization Each project begins with a Scope of Project meeting between you and our technical staff in an effort to assure the success of your project. Mice are then immunized according to the schedule below and titer is monitored by ELISA until sufficient titer is reached to warrant fusion between mouse spleen cells and an immortal, non-secreting myeloma cell line.
Cloning Up to three potential positive cell lines from Phase II will be expanded to produce sufficient culture to preserve the parent cell line and to reclone the identified positives by limiting dilution. The expansion culture of the three selected positive cell lines will be preserved in liquid nitrogen. Up to seventeen additional positive cell lines may be cryopreserved shipped to you.
Recloning of selected positives will be performed twice to evaluate antibody production stability. Clones must exhibit >95% antigen-specific antibody production to be considered stable
Deliverables At the end of the project the resultant cell line is delivered to, and is the property of, the client. As a safety measure Crown Bio can hold, store, and maintain daughter cell lines, which are available to the client at any time.
